A three trailer road train rollover has completely blocked the Stuart Highway three and a half kilometres north of Pine Creek.
Territory Duty Superintendent Lee Morgan said the single vehicle crash was reported to police at 2:30am this morning, Northern Territory Police say.
"The road train was heading south when it rolled, the cab and one trailer have blocked both north and south lanes.
"Inclement weather in the area have made the possibility of a diversion impossible," he said.
"A heavy lift crane is heading to the scene from Darwin, it is anticipated the highway will remain blocked, possibly until midday."
Two males were taken to the Pine Creek Clinic for assessment.
