About 3.45pm, white and red Freightliner truck towing trailers, was travelling northbound on Bussell Highway, near Harmans Mill Road.
The trailer of the Freightliner truck was involved in a collision with two vehicles travelling southbound - a black Volkswagen Tiguan station sedan, and a gold Subaru Outback station sedan.
Emergency services attended the scene.
The 53-year-old female driver of the gold Subaru was airlifted to Royal Perth Hospital, but later died as a result of critical injuries received.
The 28-year-old female driver of the black Volkswagen hatchback was conveyed to Busselton Hospital for treatment of non-life-threatening injuries.
The 56-year-old male driver of the Freightliner stopped at the scene, and was uninjured.
